Abstract
To determine how well protein secondary-structure estimates obtained from particle beam spectra correlate with estimates obtained from other IR sampling methods, a series of globular protein spectra was acquired. In addition to solid film deposit produced by the particle beam, spectra were obtained by the Attenuated Total Reflectance (ATR) Circle cellTM, on ZnSe as evaporated films, and with Diffuse Reflectance in KCl. The proteins used in this study were classified into types based on relative amounts of secondary structures as determined by FT-IR spectrometry. Estimates of secondary structure content from the solid- state deposits prepared with particle beam, evaporated films, and ATR spectra were very similar. The harsh environment required for diffuse reflectance measurements may explain why estimates by this method did not compare well with the other techniques employed.
